Recent studies have elucidated key enzymes and regulatory networks governing triterpenoid saponin biosynthesis. One investigation employed methyl jasmonate elicitation and full-length transcriptome profiling in Saponaria vaccaria to identify enzymes catalysing oxidation and glycosylation steps, including a cellulose synthase-like glucuronyltransferase and novel UDP-sugar modifying enzymes. This work revealed the biosynthetic route to rare sugar donors and set the stage for synthetic biology production of oleanane-type saponins. Another study sequenced the genome of soapwort (Saponaria officinalis) and, through genome mining and combinatorial expression, characterised 14 enzymes completing the pathway to saponarioside B, an oleanane saponin with adjuvant potential. The discovery of a cytosolic glycoside hydrolase transglycosidase for quinovose addition highlights noncanonical mechanisms for sugar attachment, opening avenues for engineering immunostimulant and drug-delivery agents.
