Summary

Saponins are a diverse class of natural glycosides characterised by amphiphilic structures comprising a hydrophobic aglycone (sapogenin) and one or more hydrophilic sugar chains. They fall into two principal categories—triterpenoid and steroidal—according to the nature of the sapogenin backbone. Biosynthesis proceeds via the mevalonate and MEP pathways in plant cells, giving rise to extensive structural variation through differing aglycone cores, sugar types and linkages. In botanical systems, saponins contribute to defence against pathogens and herbivores by perturbing membrane integrity, and they also modulate symbiotic interactions. Analytical advances in mass spectrometry, nuclear magnetic resonance and chromatographic separation have accelerated the characterisation of novel saponins, revealing subtle stereochemical and glycosidic diversity. Bioactivity studies indicate pronounced anti-inflammatory, antimicrobial, anticancer and adjuvant properties, fostering applications ranging from herbal anti-inflammatories and nutraceuticals to bioherbicides and vaccine adjuvants. Global demand for sustainable agrochemicals and pharmacological leads has focused attention on saponin-rich genera such as Agave, Panax and Dioscorea, highlighting both the ecological roles and translational potential of these compounds.

Triterpenoid saponin: A saponin subclass featuring a sapogenin based on a 30-carbon triterpene skeleton.

Steroidal saponin: A saponin subclass featuring a sapogenin with the characteristic four-ring steroid framework.
