Sertoli Cell Biology and Spermatogenic Regulation
Summary
Sertoli cells are the foundational somatic component of the seminiferous epithelium, orchestrating the progression of spermatogonia to mature spermatozoa. These cells establish a specialised microenvironment through the formation of the blood–testis barrier, which segregates germ cells from systemic circulation and maintains an immune-privileged site. Sertoli cells supply developing germ cells with metabolic substrates, trophic factors and structural support, while dynamically responding to endocrine cues—especially follicle-stimulating hormone and testosterone—to regulate proliferation, differentiation and apoptosis within the seminiferous tubules. Their tight junctions and adherens junctions not only facilitate germ-Sertoli cell contact but also modulate paracrine signalling pathways that underpin the self-renewal of spermatogonial stem cells. Beyond physical scaffolding, Sertoli cells secrete immunomodulatory and growth factors that influence Leydig cell function and systemic hormonal feedback. Dysregulation of Sertoli cell physiology by genetic, environmental or stress-related factors can compromise barrier integrity, alter the transcriptome of germ cells and lead to subfertility or infertility. Recent advances in in vitro modelling, transcriptomic profiling and mathematical simulation continue to elucidate the molecular networks by which Sertoli cells coordinate spermatogenesis and preserve male reproductive health.

The graph below shows the total number of articles in sertoli cell biology and spermatogenic regulation across all publications each year (not limited to Nature Index journals).
Technical terms
Sertoli cell: A somatic ‘nurse’ cell in the seminiferous epithelium that supports germ cell development and maintains the blood–testis barrier.
Spermatogenesis: The multistage process by which spermatogonial stem cells undergo mitosis, meiosis and differentiation into mature spermatozoa.
Blood–testis barrier: A specialised junctional complex between adjacent Sertoli cells that creates a microenvironment for germ cell development and protects against immune attack.
Follicle-stimulating hormone (FSH): A pituitary hormone that binds to receptors on Sertoli cells to stimulate cAMP-dependent pathways, protein synthesis and support of spermatogenesis.
Tight junction: A cell–cell adhesion structure between Sertoli cells critical for barrier integrity and selective transport of molecules into the seminiferous compartment.
Transcriptome: The complete set of RNA transcripts produced by a cell, reflecting its gene-expression profile at a given developmental stage or in response to stimuli.

About these summaries
This Nature Research Intelligence Topic summary is created with the cited references and a large language model. We take care to ground generated text with facts, and have systems in place to gain human feedback on the overall quality of the process in line with our AI principles. We strive to create accurate and useful summaries for people unfamiliar with the research topic and that supports this goal. These pages are a beta release and will be updated as we learn how best to help people gain value from a research topic summary.
