Sertoli Cell Dynamics in Male Reproductive Biology
Summary
Sertoli cells are the essential somatic component of the seminiferous epithelium, orchestrating spermatogenesis by providing physical support, nutrient supply and regulatory cues to developing germ cells. Their dynamic cytoskeletal architecture underpins the formation and remodelling of specialised junctions—most notably the blood-testis barrier (BTB) and ectoplasmic specialisations—that compartmentalise the testis and facilitate germ cell movement. Throughout the epithelial cycle, coordinated signalling pathways regulate actin and microtubule networks in Sertoli cells to allow sequential opening and resealing of junctions, ensuring both the immune privilege of germ cells and efficient progression from spermatogonia to mature spermatozoa. Aberrations in these dynamics are implicated in male infertility, making Sertoli cell biology a focal point for understanding and potentially treating reproductive disorders. Recent advances have illuminated post-transcriptional control, polarity complex function and intracellular kinase cascades as central determinants of Sertoli cell behaviour, highlighting their global significance and furnishing new targets for therapeutic intervention.

Recent studies have identified the role of Long-form collapsin response mediator protein-1 (LCRMP-1) in maintaining F-actin organisation through modulation of PI3K-Akt signalling to facilitate spermiation and support spermatid release. These investigations in Lcrmp-1 knockout mice reveal stage-dependent expression of LCRMP-1 in the seminiferous epithelium and its necessity for cytoskeletal homeostasis and fertility. Additionally, foundational work on the polarity protein Crumbs homolog-3 (CRB3) has elucidated its regulation of ectoplasmic specialization dynamics by organising actin bundles at the blood-testis barrier, thereby coordinating Sertoli cell–spermatid adhesion and transport.
