Summary
Short-chain fatty acids (SCFAs)—principally acetate, propionate and butyrate—are microbial metabolites generated by the fermentation of dietary fibre and resistant starch in the colon. These small organic acids act locally to strengthen gut barrier function and modulate immune tone, and systemically to influence energy harvest, lipid metabolism, glucose homeostasis and appetite. SCFAs engage specific G-protein-coupled receptors on enteroendocrine and immune cells, trigger the secretion of gut hormones such as peptide YY (PYY) and glucagon-like peptide-1 (GLP-1), and activate intracellular sensors like AMP-activated protein kinase (AMPK). Together, these mechanisms promote fatty-acid oxidation, inhibit de novo lipogenesis, enhance insulin sensitivity and suppress food intake. Dysregulation of SCFA production and signalling has been implicated in obesity, type 2 diabetes, non-alcoholic fatty liver disease and cardiovascular risk. In light of growing global metabolic disease burden, there is intense interest in dietary and microbial interventions—ranging from fermentable-fibre supplementation to targeted probiotic and postbiotic strategies—that selectively boost SCFA output or mimic their effects, with the aim of restoring metabolic health.

Short-chain fatty acids (SCFAs): Saturated aliphatic acids with fewer than six carbons, produced by microbial fermentation of non-digestible carbohydrates in the colon.
G-protein-coupled receptors (GPCRs): Cell-surface receptors, including FFAR2 and FFAR3, that bind SCFAs and initiate intracellular signalling affecting metabolism and immunity.
Glucagon-like peptide-1 (GLP-1): An incretin hormone released by enteroendocrine L cells that enhances insulin secretion and contributes to satiety.
Peptide YY (PYY): A gut hormone co-secreted with GLP-1 that slows gastric emptying and reduces appetite.
AMP-activated protein kinase (AMPK): A master cellular energy sensor activated by SCFAs that promotes energy expenditure and inhibits lipid biosynthesis.
