Conversely, poor sleep can accelerate neurodegenerative processes, contributing to declines in memory, executive function and global cognition. A bidirectional relationship exists whereby chronic disease burdens further disrupt sleep, creating a cycle that heightens frailty and functional impairment. Understanding the mechanistic underpinnings—from synaptic homeostasis to systemic inflammation—has become central to designing effective screening, prevention and intervention strategies. Population ageing worldwide underscores the urgency of identifying modifiable sleep parameters to promote healthy longevity and reduce the societal burden of age-related conditions.

Recent studies have applied multivariate neuroimaging methods to link composite sleep-health dimensions with distributed functional connectivity. One investigation using partial least squares analysis revealed a brain connectome biomarker—centred on attentional and thalamic networks—that covaries with sleep health and shows promise for early detection of insomnia and related cognitive risk.
Another large-scale analysis of actigraphy data in older cohorts has characterised how sleep fragmentation and reduced sleep efficiency forecast accelerated cognitive decline. By integrating wearable-monitoring with longitudinal follow-up, researchers have delineated age-specific thresholds at which sleep disruption portends increased incidence of mild cognitive impairment over a five-year period.
