Summary
Social cognition encompasses the mental operations underlying social interactions, including perception, interpretation and response to the intentions, emotions and behaviours of others. In autism spectrum disorders (ASD), disruptions in these processes manifest as atypical eye contact, diminished social motivation, and altered reward valuation of social stimuli. Contemporary research reveals that both cortical and subcortical networks contribute to these alterations. Cortically, atypical functional connectivity in medial prefrontal and temporal regions undermines theory of mind and self-referential processing. Subcortically, aberrant activation in structures such as the amygdala, superior colliculus and periaqueductal grey shapes early perception of social cues. Moreover, dopaminergic circuits in the ventral striatum display attenuated anticipatory responses to social rewards, underpinning reduced social seeking. Integrative models now emphasise the interplay between sensory processing anomalies, motivational deficits and atypical learning through social channels. Understanding these intertwined mechanisms is essential for devising interventions that target neural circuitry, enhance social reward salience and harness learning via social interaction.

Advanced digital phenotyping in paediatric settings has demonstrated that computer vision and machine learning can reliably detect early behavioural markers of ASD within routine well-child visits. An app presents stimuli designed to elicit gaze and gesture patterns, and an algorithmic fusion of multiple digital behavioural signatures achieved high sensitivity and specificity across diverse demographic groups. This work highlights the potential for objective, scalable screening tools that reduce reliance on subjective questionnaires and may mitigate disparities in early diagnosis.
Investigations of subcortical face-processing pathways have revealed that constraining gaze to the eye region provokes hyperactivation throughout the superior colliculus–pulvinar–amygdala circuit in autistic adults. Such heightened responses are proposed to drive compensatory avoidance of eye contact in daily life. These findings underscore the role of early, automatic neural responses in shaping subsequent social behaviour and the developmental tuning of cortical face regions.
