Summary
Sperm motility is a finely tuned process essential for successful fertilisation, relying on the interplay of ion fluxes, energy metabolism and signal transduction pathways. Motility initiation and maintenance involve the coordinated activity of ion channels and transporters that regulate intracellular calcium, bicarbonate and proton concentrations. These ionic changes activate soluble adenylate cyclase to produce cyclic AMP, which in turn engages protein kinase cascades such as AMP-activated protein kinase and protein kinase A to modulate flagellar beat frequency and waveform. Concurrently, energy supply through glycolysis and mitochondrial respiration ensures sustained dynein ATPase activity along the axoneme. Protein kinases sensitive to calcium/calmodulin further refine motile behaviour, while Ca2+/Mg2+-ATPases restore ionic homeostasis. Hormonal and paracrine factors present in the male reproductive tract, including steroidal and prostaglandin mediators, influence these intracellular pathways to optimise sperm function. Dysregulation at any node—ion transport, second-messenger generation or kinase activity—can culminate in reduced fertilising potential, highlighting both the complexity of motility control and its significance for male fertility and contraceptive development.
