Summary

Stathmin (STMN1) is a cytoplasmic phosphoprotein that regulates microtubule dynamics by binding tubulin dimers and promoting their depolymerisation. Aberrant upregulation of stathmin has been reported across a spectrum of malignancies, where it drives tumour cell proliferation, motility and metastatic potential. Elevated stathmin expression often correlates with advanced tumour stage, lymph node involvement and reduced overall or disease-free survival, positioning it as an independent prognostic biomarker. Beyond its role in cell division, stathmin modulates response to anti-microtubule agents such as paclitaxel and vincristine, thus serving as a potential predictive marker for chemotherapy efficacy. Mechanistic studies have further revealed that stathmin interfaces with key signalling pathways, including p53, MAPK and PI3K/Akt, and is regulated by non-coding RNAs. Taken together, the global significance of stathmin encompasses both its diagnostic and therapeutic dimensions, highlighting its value in risk stratification and as a target for novel molecular interventions.

A foundational study investigated STMN1 in gallbladder carcinoma, revealing markedly higher expression in tumour tissues relative to normal mucosa. Functional assays demonstrated that STMN1 silencing impaired proliferation, induced apoptosis and delayed G2/M transition in vitro. In mouse xenograft models, knockdown of STMN1 significantly suppressed tumour growth, implicating its involvement in p38 MAPK and p53/p21 signalling cascades. These findings support STMN1 as a driver of gallbladder carcinoma progression and underscore its promise as a therapeutic target to disrupt microtubule stability in aggressive malignancies.
