Summary

Syndecans are a family of transmembrane heparan sulfate proteoglycans that serve as versatile co-receptors for growth factors, chemokines and extracellular matrix (ECM) components. In tumour microenvironments, syndecans modulate key processes such as angiogenesis, immune cell recruitment, cell–matrix adhesion and mechanotransduction. Their extracellular domain binds heparan sulfate chains, concentrating soluble ligands and presenting them to canonical receptors, while the short cytoplasmic tail interacts with intracellular adaptors to coordinate cytoskeletal remodelling and endocytic trafficking.
