Synergistic Interactions in Natural Product Pharmacology
Summary
Natural products frequently comprise complex mixtures of bioactive compounds whose combined effects can exceed those of individual constituents. Synergistic interactions arise when two or more components act in concert to enhance efficacy, reduce toxicity or circumvent resistance. Such phenomena have been observed across antimicrobial, anticancer, anti‐inflammatory and metabolic applications. Mechanisms include pharmacodynamic synergy, whereby complementary targets or pathways are co-modulated, and pharmacokinetic modulation, in which one constituent alters absorption, distribution, metabolism or excretion of another. Advances in systems biology, metabolomics and network pharmacology are now enabling deconvolution of these multi-component effects, while novel in vitro and in silico platforms permit rapid screening of combinations. Despite methodological challenges in design, quantification and standardisation, elucidating synergistic relationships in plant extracts, essential oils and herbal formulations holds promise for more effective and safer therapeutics. This synergy‐centred paradigm is of global significance, offering routes to combat antimicrobial resistance, optimise cancer regimens and develop affordable medicines in resource-limited settings.
