Tyrosine Kinase Signaling in Cytokine Pathways
Summary
Tyrosine kinase signalling underpins the cellular response to a myriad of cytokines that regulate immunity, haematopoiesis and tissue homeostasis. Engagement of a cytokine with its cognate receptor induces conformational changes and often receptor dimerisation or higher‐order oligomerisation. This juxtaposition brings associated cytoplasmic tyrosine kinases, principally members of the Janus kinase (JAK) family, into proximity, where they engage in reciprocal trans‐phosphorylation. Activated JAKs phosphorylate specific tyrosine residues within receptor intracellular domains, creating docking sites for downstream effectors such as signal transducers and activators of transcription (STATs). Phosphorylated STATs dimerise and translocate to the nucleus to initiate context‐dependent gene programmes. Fine tuning of this cascade is achieved by negative regulators including suppressors of cytokine signalling (SOCS), protein tyrosine phosphatases and regulatory adaptor proteins. Beyond the core JAK–STAT axis, cross‐talk with mitogen-activated protein kinase, phosphatidylinositol 3-kinase/Akt and nuclear factor-κB pathways expands the scope of biological outcomes. Aberrant tyrosine kinase signalling in cytokine pathways contributes to immunodeficiency, chronic inflammation and oncogenesis, and small-molecule inhibitors targeting JAKs have emerged as transformative therapies in autoimmune disease and haematological malignancies.
