Summary
Reptiles present unique challenges for anaesthesia and pain management owing to their ectothermic metabolism, variable cardiovascular responses and distinctive renal portal system. Effective protocols must account for temperature‐dependent drug clearance and species‐specific respiratory adaptations. Injectable agents such as alfaxalone, ketamine and α2-adrenergic agonists are commonly combined to achieve sedation, muscle relaxation and analgesia, while inhalant anaesthetics like isoflurane provide adjustable maintenance. Analgesic strategies draw on opioids, non-steroidal anti-inflammatory drugs (NSAIDs) and local anaesthetics to control nociception and modulate inflammatory processes. Monitoring relies on behavioural endpoints (righting reflex, limb withdrawal), cardiovascular parameters (heart rate, blood pressure) and temperature regulation. Recent advances emphasise multimodal approaches that minimise cardiorespiratory depression and leverage biomarkers—such as circulating microRNAs—for objective assessment of surgical stress and pain. Continued refinement of protocols is essential for enhancing welfare in captive, research and conservation settings worldwide.

By sampling plasma before and after surgery, researchers identified distinct miRNA signatures associated with acute surgical stress. Two candidate miRNAs demonstrated strong diagnostic potential for discriminating between pre- and early post-operative states, suggesting a novel, minimally invasive biomarker approach to gauge analgesic efficacy and recovery in chelonian medicine.
