Transient receptor potential canonical 3 is required for HPV-induced malignant transformation of cervical epithelial cells

This study reveals that the TRPC3 channel is essential for HPV-driven cervical cancer. TRPC3 promotes malignant transformation by regulating the RASA4/MAPK pathway and EMT. Targeting TRPC3 could be a new therapeutic strategy.

Ubiquitination of RIPK1 regulates its activation mediated by TNFR1 and TLRs signaling in distinct manners

RIPK1 is a critical kinase which mediates necroptosis, apoptosis and inflammation. Regulation of RIPK1 by ubiquitination is being intensively investigated. Here, the authors made knock-in RIPK1-K612R mice and demonstrate that this mutation alters the RIPK1 ubiquitinylation pattern and inhibits its prodeath kinase activity in response to TNFα but sensitizes cell death to TLRs signals.

MHC-I mediated antigen presentation is an important element of the anti-tumour immune response. Here authors identify a tumour immune escape mechanism by which the cancer cells express the ubiquitin E3 ligase UHRF1 in the cytoplasm instead of the nuclear expression pattern observed in normal tissues, and this results in degradation of MHC-I and thus diminished antigen presentation and anti-tumour T cell response.

Atomic-scale strain engineering of atomically resolved Pt clusters transcending natural enzymes

Strain engineering is promising for improving catalytic capability of biocatalysts, but it is challenging to modify the atomic-scale strain for specific enzyme-like reactions. Targeted bisulfite sequencing reveals changes in DNA methylation associated with nuclear reprogramming

Although technically feasible, whole-genome analysis of cytosine methylation using bisulfite sequencing remains prohibitively expensive for large eukaryotic genomes. Deng et al. use 30,000 nondegenerate padlock probes to capture ∼66,000 bisulfite-converted sites in human CpG islands and compare their methylation in fibroblasts, embryonic stem cells and induced pluripotent stem cells.

A multilaboratory analysis characterized the ability of 27 different labs to identify 20 proteins at equimolar concentrations in a highly purified test sample mixture using mass spectrometry.
