FBXL5 deficiency in CD11c+ myeloid cells causes iron overload, enhancing NF-κB recruitment to the IL-6 promoter and increasing IL-6 production. This exacerbates neutrophil infiltration and liver injury in mice, while IL-6 neutralization mitigates damage, revealing iron regulation as a therapeutic target.
