Summary
Eucalyptol, also known as 1,8-cineole, is a naturally occurring monoterpene oxide extracted chiefly from eucalyptus and other aromatic plants. Its pharmacological profile encompasses mucolytic, bronchodilatory and anti-inflammatory activities that have underpinned traditional and modern therapeutic uses in airway disorders such as asthma, chronic obstructive pulmonary disease (COPD), bronchitis and rhinosinusitis. Mechanistically, eucalyptol modulates innate immune signalling by attenuating pro-inflammatory mediators, including tumour necrosis factor-α, interleukins and nitric oxide, through downregulation of transcription factors and kinases such as NF-κB and p38 MAPK. It also interferes with pattern recognition receptor pathways and inflammasome activation, thereby dampening exaggerated inflammatory responses without compromising host defence. Clinically, adjunctive administration has been shown to reduce mucus viscosity, enhance airway clearance and lower exacerbation rates, while its favourable safety profile and compatibility with standard bronchodilators and corticosteroids point to broader applications in chronic respiratory care. Ongoing work seeks to optimise delivery systems and dosage regimens to maximise pulmonary deposition and sustain therapeutic concentrations in the airways.
