Metformin inhibits nuclear egress of chromatin fragments in senescence and aging

Kumazawa, Xu, Wang and colleagues identify nuclear egress as the mechanism by which cytoplasmic chromatin fragments relocate from the nucleus to the cytoplasm in senescence. They link inflammation to metabolism by showing this egress is regulated by AMPK and metformin.

p53 enhances DNA repair and suppresses cytoplasmic chromatin fragments and inflammation in senescent cells

The transfer of nuclear chromatin to the cytoplasm drives the pro-aging inflammatory phenotype of senescent cells. Here, Miller et al. show that this process is suppressed by p53 and that activation of p53 reverses some features of aging in vivo.
