Pulmonary fibrosis is a progressive lung disease characterized by scarring and impaired function, with vascular endothelial cells playing a key role in disease progression. Here, the authors show that endothelial mechanosensitive PIEZO1 promotes fibrosis by activating the CAPN2-STAT3-IL-33 signaling axis, identifying a promising therapeutic target for treatment.
