Dearomatization offers a route towards three dimensional spirocyclic architectures, which are essential in bioactive molecules and pharmaceuticals, but biocatalytic dearomatization for obtaining these spirocyclic frameworks is underexplored. Here, the authors report a photoenzymatic platform synergizing visible-light activation with engineered nicotinamide-dependent ketoreductase catalysis to achieve highly enantioselective dearomative spirocyclization of N-hydroxyphthalimide esters.
