Controlling site selectivity in carbonyl desaturation to prepare α,β-unsaturated carbonyl compounds is challenging. Now, engineered ‘ene’-reductases are shown to enable the biocatalytic desaturation of cyclic ketones to enones with high site divergence. This platform allows late-stage functionalization of complex molecules, including terpenoids, and provides access to chiral enones via desaturative kinetic resolution.
